We are currently focusing now on identifying the molecular basis that provides specific and selective interactions within macromolecular signaling complexes and is disrupted in diseases. This approach will help us towards structure-based drug design.

X-ray crystallography approach

PKA RIb holoenzyme structure

Dr. Ronit Ilouz was the first to solve the X-ray crystal structure of PKA RIb holoenzyme where all the domains are visualized. The full length crystal structure consists of two catalytic subunits bound to a regulatory subunit dimer. This work changed our understanding of PKA allosteric properties and and isoform specificity that was not anticipated without the complete structure.
